研究方向聚焦于基于定量系统药理学与多尺度电生理技术的中药抗心律失常有效组分群发现与机制解析。针对中药复方“多组分-多靶点”协同作用中量效关系不清的核心难题,您以临床验方心速宁胶囊和活心丸为研究对象,致力于揭示其治疗心房颤动、室性早搏、缺血再灌注损伤及心力衰竭的科学内涵。研究采用“计算预测+实验验证”的闭环策略:一方面利用自建的复方组分定量协同算法(CM-QSP)、分子动力学模拟及心肌动作电位仿真模型,从海量组合中精准筛选最优组分配伍;另一方面结合高通量膜片钳、iPSC诱导分化心肌细胞、人工心肌组织、离体心脏灌流及在体动物模型,在离子通道、细胞、组织、器官及整体多个尺度上验证药效。核心目标是阐明中药多组分通过协同调控心脏多离子通道平衡、氧化应激、线粒体功能及钙稳态等网络机制,从而安全有效治疗心律失常的分子机理;并据此挖掘出化学组成明确、比例固定、疗效优于原复方的“有效组分群”(即现代复方化学药)。该研究旨在打破传统中药“黑箱”,构建中药复方现代化研究新范式,推动心血管中药从“经验用药”向“精准设计”转化,为抗心律失常创新药物研发提供坚实的理论基础与技术支撑。
